I just installed TotalMounter Pro v2.0.1000.1058, and I can't create a virtual burner. I select "Create a new file, size:", choose Disc Type of "DVD / Blue-Ray (sic)", set the image size in MBs to 4700, choose a file path on my hard drive ("C:\Users\ecto\img\test.iso" [which does not exist]), and then click "Finish." I expect the program to create test.iso, but instead I get a single, cryptically terse popup message,"No device left!"
Is it because the image doesn't yet exist?

I think the problem you are facing is no driver connected, when you install the software, there should be a message to ask you install driver, did you accept and install the driver?
And also, if you are not running under the administrator's permission, please run it as administrator and try again.

For me, unmounting something I'd mounted always caused the problem. Unmounting makes the extra drive disappear and any future attempts to mount gives "No device left" error.
The solution for me is to restart my computer. The extra drive comes back and you can mount again.
